While the original comic series didn’t offer a definitive explanation of what caused the plague to wipe out all living mammals with Y chromosomes — except Yorick, his monkey amps, and Dr. Sonmori — it provided various theories.
What does series writer Brian Vaughan have to say about the cause of the plague?
For his part, author Brian Vaughan someone once said: “I like that people don’t necessarily know what it is. In interviews, we always say we’ll tell people what actually caused the plague. The problem is, we never say when to say it.”

Dr. Matsumori’s theory hold The Y chromosome has been “reasonably self-destructing for hundreds of millions of years.” The birth of his first successful clone “triggered a ticking time bomb that had been ticking for thousands of years”.
The theory behind the Setauket Ring has to do with the removal of a sacred amulet from the Jordanian state, exactly what Agent 355 is played by Ashley Romans in the TV adaptation.
Various other theories have been proposed by other figures. For example, according to air traffic controllers, there was an ecstatic event that punished women for their original sin by eliminating men.
